Chromogenic substrates are colorless, soluble molecules composed of two key components:
- A chromophore – a chemical group that, upon enzymatic cleavage, releases color.
- A specific enzymatic substrate, typically a digestible carbohydrate.
These compounds serve as valuable tools for enzymatic detection because they specifically bind to the target enzyme. When this reaction occurs, the enzyme catalyzes the separation of the chromophore group, producing an insoluble product with a distinct color that confirms the presence and activity of the enzyme being studied. The intensity of the color can be quantified in chromogenic assays used in techniques like Western blotting, ELISA, immunohistochemistry, enzymatic assays, and microbial detection in culture media.
