CytoQuestTM CR - a non-invasive separation and recovery system for circulating rare cells

Abnova CytoQuest™ CR——针对循环罕有细胞非侵袭式的分离回收系统 Molecular biology and clinical studies have shown that tumor invasion and micrometastasis are likely to occur early in tumorigenesis. At present, the discovery and diagnosis of clinical tumors are still highly dependent on imaging and serum tumor markers, and it is difficult to detect tumor metastasis or recurrence early, and it is difficult to reflect the efficacy in time. Therefore, early detection of micrometastasis is not only important for the judgment of tumor recurrence and prognosis, but also valuable for guiding clinical treatment. Detection of circulating tumor cells (CTCs) is helpful for early detection of micrometastasis in tumors, monitoring postoperative recurrence, assessing efficacy and prognosis, and selecting appropriate individualized treatments. As early as 1869, Australian scientist Thomas Ashworth proposed the concept of CTCs. In recent years, with the in-depth study of tumor metastasis mechanisms and the continuous improvement of detection techniques, some breakthroughs have been made in the detection of CTCs.

CytoQuestTM CR
is a non-invasive, cyclic rare cell sorting system developed by Abnova and the UCLA (University of California Los Angeles) Nano Research Institute. Based on molecular affinity, the CytoQuestTM CR system uses a temperature-sensitive array of nanopillars to capture, count, separate, and recover Circulating Rare Cells (CRCs) from a sample in a non-invasive manner. At present, in translation studies and clinical research, CRCs mainly include circulating tumor cells (CTCs), circulating stem cells (CSCs), and circulating fetal cells (CFCs).

Since the proportion of CTCs in the blood is very small (one hundred parts per million), there is still a problem that needs to be overcome for the enrichment of CTCs. At present, CTCs enrichment methods mainly include immunomagnetic separation method and morphological-based enrichment method. The former has serious white blood cell contamination problems, while the latter is not defined because of the size of tumor cells, so its separation sensitivity is questioned. CTCs can also be obtained by flow sorting, but the cells are killed due to various external forces that the cells are subjected to in the flow sorting system, and there is no activity. How to effectively collect and separate high-purity, dynamic CTCs has become a major challenge!

CytoQuestTM CR utilizes immobilized antibodies, nanotechnology chips, and TCx® thermal sensing technology to achieve cell sorting. At 37 degrees, the antibody linked to the nanocolumn chip is exposed to capture the cells. At 4 degrees, the antibody is automatically concealed to release the cells (as shown in the figure below). The CTCs enriched by the system are less damaged and more active. Because CytoQuestTM CR chip antibodies are flexible and interchangeable, and there is no morphological size requirement for the cells sorted by themselves, the system is not only for CTCs, but is widely used for sorting other rare cells such as circulating stem cells (CSCs), circulation. Fetal cells (CFCs), etc.

The CRCs captured on the chip can be counted under the microscope in combination with fluorescent staining methods, or combined with a clinical microdisc to achieve true single cell separation.

CytoQuestTM CR provides high-purity, high-survival circulating rare cells (CRCs) that can be followed by numerous studies, which are summarized below:

  • Qualitative analysis of proteins: immunofluorescence staining, ortho-ligation analysis
  • Genetic analysis: fluorescence in situ hybridization (FISH), copy number variation analysis (CNV), single point mutation analysis (SNP), NGS sequencing
  • Cell detection: primary cell line construction, compound screening

Superior advantages of the CytoQuestTM CR system :

  • Integrated fluid, pump and temperature control system
  • Affinity circulating tumor cell capture and temperature-sensitive release
  • Simple circulating tumor cell capture and counting function (optional)
  • Touch screen and friendly user interface

The Cell Count of the CytoQuestTM CR System is currently the only FDA-approved product that can be used as a diagnostic indicator for clinical breast cancer, prostate cancer, and bowel cancer. For translation and clinical medical research, CytoQuestTM CR is a powerful aid that helps identify and track cancer cell changes and metastasis, opening up new horizons in clinical diagnostics and pharmaceuticals.
